چکیده

Background Proteolytic enzymes within venoms from different scorpion species belong to serine and metalloproteases class, and some isoforms of these enzymes show proteolytic activity over Aa and Bb subunits of fibrinogen [1-3]. Brotheas amazonicus is a non-lethal amazonian scorpion, belonging to a different group from Tityus genus, and there are scarce studies about its venom in literature [4]. Enzymes acting over fibrinogen can lead to the creation of more efficient antithrombotic drugs, and the low lethality of B. amazonicus venom is a potencial factor for developing a better drug.

Correlation between the Glycan Variations and Defibrinogenating Activities of Acutobin and Its Recombinant Glycoforms

Acutobin isolated from Deinagkistrodon acutus venom has been used to prevent or treat stroke in patients. This defibrinogenating serine protease is a 39 kDa glycoprotein containing terminal disialyl-capped N-glycans. After sialidase treatment, the enzyme showed similar catalytic activities toward chromogenic substrate, and cleaved the Aα chain of fibrinogen as efficiently as the native acutobin...

Evidence that multiple proteases of Bacillus subtilis can degrade fibrin and fibrinogen

Fibrinolytic enzyme produced by Bacillus spp. are known to possess an unique property to degrade fibrin blood clots. Fibrinolytic enzyme such as nattokinase has commercial applications as a therapeutic agents and functional food formulation. In this study, we report an interesting characteristic feature of Bacillus subtilis BR21, a native isolate for its ability to produce multiple proteases th...

Biochemical characterization and comparative analysis of two distinct serine proteases from Bothrops pirajai snake venom.

This study reports the isolation and biochemical characterization of two different serine proteases from Bothrops pirajai snake venom, thus providing a comparative analysis of the enzymes. The isolation process consisted of three consecutive chromatographic steps (Sephacryl S-200, Benzamidine Sepharose and C2/C18), resulting in two serine proteases, named BpirSP27 and BpirSP41 after their molec...

Characterization of a fibrinolytic enzyme secreted by Bacillus amyloliquefaciens CB1 and its gene cloning.

Bacillus amyloliquefaciens CB1 was isolated from cheonggukjang, a Korean fermented soy food. B. amyloliquefaciens CB1 secretes proteases with fibrinolytic activities. A gene homologous to aprE of Bacillus subtilis, aprECB1, was cloned from B. amyloliquefaciens CB1, and DNA sequencing showed that aprECB1 can encode a prepro-type serine protease consisting of 382 amino acids. When aprECB1 was int...

Batroxobin binds fibrin with higher affinity and promotes clot expansion to a greater extent than thrombin.

Batroxobin is a thrombin-like serine protease from the venom of Bothrops atrox moojeni that clots fibrinogen. In contrast to thrombin, which releases fibrinopeptide A and B from the NH2-terminal domains of the Aα- and Bβ-chains of fibrinogen, respectively, batroxobin only releases fibrinopeptide A. Because the mechanism responsible for these differences is unknown, we compared the interactions ...
